No, VFS does not launch apps from a memplug.
VFS is a standardized way by which programmers can add external memory capability to their programs without having to write their program for each external memory adapter that comes along. This way, one set of code is written that can work with many types of memory. Such as Kopsis' Flash Adapater, Memplug SM and CF, SD/MMC, and Memory Stick.
PiDirect, however, is a means by which you can effectively "run" programs stored on the memory in your memplug. It, I assume, was written for the Memplug line of springboards only. If it were to be written as an application that supports VFS, anybody with external storage could use it, regardless of the media type.
